Identification of Transforming Growth Factor-P as a Contaminant in Factor VI11 Concentrates: A Possible Link With Immunosuppressive Effects in Hemophiliacs

چکیده
In previous studies, we have shown that some, but not all IOW-, intermediate-, and high-purity factor Vlll concentrates inhibit interleukin-2 (IL-2) secretion from phytohemagglutinin (PHA)-stimulated T lymphocytes. We now present evidence that this inhibitory action of concentrates is, at least in part, due to contamination with transforming growth factor-/3 (TGF-P). Originally identified in platelets, TGF-/3 is a 25-kD homodimer that has been shown to be a natural and potent inhibitor of many immunologic responses. Using a specific bioassay, we have measured TGF-B in various factor Vlll concentrates. While some concentrates contained substantial amounts of the cytokine, there was a wide variation in concentrations of TGF-/3 in different products. These lev-

Identification of transforming growth factor-beta as a contaminant in factor VIII concentrates: a possible link with immunosuppressive effects in hemophiliacs.
In previous studies, we have shown that some, but not all low-, intermediate-, and high-purity factor VIII concentrates inhibit interleukin-2 (IL-2) secretion from phytohemagglutinin (PHA)-stimulated T lymphocytes. We now present evidence that this inhibitory action of concentrates is, at least in part, due to contamination with transforming growth factor-beta (TGF-beta). Originally identified ...
متن کاملImmune Status of Human Immunodeficiency Virus Seropositive and Seronegative Hemophiliacs Infused for 3.5 Years With Recombinant Factor VI11
Recent studies suggest hat treatment of hemophiliacs with highly purified factor Vlll concentrates may preserve immune function. To test this hypothesis, we prospectively studied 51 hemophilic patients (21 human immunodeficiency virus [HIVI seropositive and 30 seronegative) who were on home therapy exclusively with recombinant factor Vlll (Kogenate, Miles Laboratory, Berkeley, CA) for 3.5 years...
متن کاملMaternal Serum Levels of Transforming Growth Factor ß1 (TGF-ß1) in Normal and Preeclamptic Pregnancies
Background: Successful pregnancy in allopregnant women depends upon the control of graft rejection mechanisms. It has been suggested that some immunosuppressive cytokines contribute to successful pregnancy and transplantation. Transforming growth factor beta (TGF- β) exhibits potent immunoregulatory and anti-inflammatory properties which might prolong graft survival. Recent studies suggest a ...
متن کاملComparison of Effects of High-Fat, High-Protein and Low-Calorie Diets on Klotho Gene Expression and TGF-β Level in Serum and Liver of Old Male Wistar Rats
Background and Objectives: Different diets have important roles in human health and aging. In the current study, we aimed to characterize the Klotho, an anti-aging protein, gene expression and protein level in the serum and assess the protein levels of Transforming growth factor-β and Platelet-derived growth factor in old rats which treated with different diets (high-fat, high-protein, low calo...
متن کاملIn vitro Induction of Fetal Hemoglobin in Erythroid Cells Derived from CD133 Cells by Transforming Growth Factor-b and Stem Cell Factor
Increased fetal hemoglobin (HbF) in b-globin gene disorders ameliorates the clinical symptoms of the underlying disease. 5-azacytidine, butyrate and hydroxyurea, have been shown to activate g-globin gene expression. It has also been found that hematopoietic growth factors can influence expression of g-globin in erythroid cultures and in animal models. This study was designed to evaluate the in ...
